The second edition of GMO Myths and Truths, co-authored by genetic engineers Dr John Fagan and Dr Michael Antoniou and researcher Claire Robinson, was released on 19 May 2014 as a free online download by the sustainability and science policy platform Earth Open Source.
The second edition follows the publication two years ago of the first edition, which was downloaded 120,000 times just a few weeks after publication and was read online by several times that many visitors. The GMO debate is far from being over, as some GMO proponents claim. Instead the evidence of risk and actual harm from GM foods and crops to health and the environment has grown in the two years since we brought out the first edition.

It can be a dataset name, i.e. bacteria_odb10, or a path i.e. ./bacteria_odb10 or /home/user/bacteria_odb10. In the former case, which is the recommended usage, BUSCO will automatically download and version the corresponding dataset. In the latter case, the dataset found in the given path will be used.Lineage can be ignored if running automated lineage selection
BUSCO can obtain the last version of the lineage datasets. If the name of a dataset is passed, e.g. -l bacteria_odb10, BUSCO will download it automatically. If a full path is given to BUSCO using -l /my/own/path/bacteria_odb10, this automated management will be disabled. Otherwise, files used during the automated lineage selection process are also automatically obtained by BUSCO.
Individual datasets can also be downloaded using the busco download command, e.g. busco download bacteria_odb10. It is also possible to do a bulk download by using the following arguments in place of the dataset name: "all", "prokaryota", "eukaryota", or "virus".
If you are running BUSCO in an environment that does not have access to the Internet, you can pass the --offline argument to prevent BUSCO from attempting any download. You will have to download and unpack all files manually from -data.ezlab.org/v5/data/ and place them in the BUSCO download folder. The location of this folder should then be passed using the command line flag: --download_path /path/to/datasets.
Since the publication of the first edition of Gene Targeting: A Practical Approach in 1993 there have been many advances in gene targeting and this new edition has been thoroughly updated and rewritten to include all the major new techniques. It provides not only tried-and-tested practical protocols but detailed guidance on their use and applications. As with the previous edition Gene Targeting: A Practical Approach 2e concentrates on gene targeting in mouse ES cells, but the techniques described can be easily adapted to applications in tissue culture including those for human cells. The first chapter covers the design of gene targeting vectors for mammalian cells and describes how to distinguish random integrations from homologous recombination. It is followed by a chapter on extending conventional gene targeting manipulations by using site-specific recombination using the Cre-loxP and Flp-FRT systems to produce 'clean' germline mutations and conditionally (in)activating genes. Chapter 3 describes methods for introducing DNA into ES cells for homologous recombination, selection and screening procedures for identifying and recovering targeted cell clones, and a simple method for establishing new ES cell lines. Chapter 4 discusses the pros and cons or aggregation versus blastocyst injection to create chimeras, focusing on the technical aspects of generating aggregation chimeras and then describes some of the uses of chimeras. The next topic covered is gene trap strategies; the structure, components, design, and modification of GT vectors, the various types of GT screens, and the molecular analysis of GT integrations. The final chapter explains the use of classical genetics in gene targeting and phenotype interpretation to create mutations and elucidate gene functions. Gene Targeting: A Practical Approach 2e will therefore be of great value to all researchers studying gene function.
